Does triptolide induce lysosomal-mediated apoptosis in human breast cancer cells?

چکیده
With breast cancer plaguing the United States as the second leading cause of cancer related deaths amongst women, as well as the adverse effects of current treatment options there is a need to develop safer and noninvasive treatments. Triptolide is an extract from the herb Tripterygium wilfordii Hook F, and has been used in Chinese medicine for over two centuries and is now used to treat certain autoimmune diseases, such as rheumatoid arthritis. Based on the anti-proliferative, anti-inflammatory, and anti-cancer properties of triptolide we believe that it will stimulate apoptosis in human breast cancer cells. Triptolide is known to induce apoptosis in many cancer cells lines, but the exact mechanisms that regulate this are largely unknown. It has been suggested that triptolide activates the p53 pathway to trigger apoptosis in these cells. However, we believe that there are other mechanisms at work including the activation of lysosomal-mediated apoptosis.

Triptolide induces lysosomal-mediated programmed cell death in MCF-7 breast cancer cells
BACKGROUND Breast cancer is a major cause of death; in fact, it is the most common type, in order of the number of global deaths, of cancer in women worldwide. This research seeks to investigate how triptolide, an extract from the Chinese herb Tripterygium wilfordii Hook F, induces apoptosis in MCF-7 human breast cancer cells. Accumulating evidence suggests a role for lysosomal proteases in the...

Aim: Bcl-2 is a potential target for tumor treatment. The inhibition of the Bcl-2 production is research target of attract in the field of anti-cancer drug development. Recently, the assessment of antitumor activity appeared to be promising for pyrazole derivatives. Therefore, this study was designed to investigate the anti-cancer effects of novel pyrazole derivatives (HN1and HN2.). Material an...
متن کاملTriptolide has anticancer and chemosensitization effects by down-regulating Akt activation through the MDM2/REST pathway in human breast cancer
Triptolide has been shown to exhibit anticancer activity. However, its mechanism of action is not clearly defined. Herein we report a novel signaling pathway, MDM2/Akt, is involved in the anticancer mechanism of triptolide. We observed that triptolide inhibits MDM2 expression in human breast cancer cells with either wild-type or mutant p53. This MDM2 inhibition resulted in decreased Akt activat...
متن کاملInhibition of tumor cellular proteasome activity by triptolide extracted from the Chinese medicinal plant 'thunder god vine'.
AIMS The molecular mechanisms of triptolide responsible for its antitumor properties are not yet fully understood. The ubiquitin/proteasome system is an important pathway of protein degradation in cells. This study investigated whether triptolide may inhibit proteasomal activity and induce apoptosis in human cancer cells. MATERIALS AND METHODS In vitro proteasome inhibition was measured by in...
متن کاملThe Probiotic Bacteria Induce Apoptosis in Breast and Colon Cancer Cells: An Immunostimulatory Effect
Background: Uncontrolled cell proliferation and resistance to apoptosis are the main characteristics of cancer cells. Therefore, a substance with the capability to induce apoptosis in cancer cells could be known as an anti-cancer material. Probiotics are useful microorganisms that are crucial for the host’s health.Materials and Methods: In the present stu...
